RAMAN SPECTROSCOPY AND DIFFUSE REFLECTANCE SPECTROSCOPY FORDIAGNOSIS OF HUMAN CANCER AND ACANTHOSIS NIGRICANSby SUNEETHA DEVPURAAugust 2012Advisor : Dr. Ratna NaikCo-Advisor : Dr. Steven J. Most common types of cancers in adultsinclude prostate, lung, breast, colorectal and head and neck squamous cell carcinoma, whileamong children; leukemia, and brain and central nervous system cancers are quitecommon. In each of these cases, early detection of the cancer or disease dramaticallyincreases the chances of successful treatment. In recent years, there has been muchinterest in using Raman spectroscopy and diffuse reflectance spectroscopy as analytical optical spectroscopic methods for early diagnosis of diseases. Raman spectroscopy can beused to measure changes in the bio-molecular composition of a tissue specimen, and diffusereflectance spectroscopy can measure chromophores of the skin. In this research, archived(formalin-fixed paraffin processed) tissues of head and neck squamous cell carcinoma, prostate, and pediatric tumors have been investigated using Raman spectroscopy. We haveutilized statistical methods such as principal component analysis (PCA) and discriminantfunction analysis (DFA) to analyze the spectral output and distinguish between normal and

Critical comparison of diffuse reflectance spectroscopy and colorimetry as dermatological diagnostic tools for acanthosis nigricans: a chemometric approach

Quantification of skin changes due to acanthosis nigricans (AN), a disorder common among insulin-resistant diabetic and obese individuals, was investigated using two optical techniques: diffuse reflectance spectroscopy (DRS) and colorimetry. Measurements were obtained from AN lesions on the neck and two control sites of eight AN patients. A principal component/discriminant function analysis suc...

Quantitative skin color measurements in acanthosis nigricans patients: colorimetry and diffuse reflectance spectroscopy.

Tristimulus colorimetry and diffuse reflectance spectroscopy (DRS) are white-light skin reflectance techniques used to measure the intensity of skin pigmentation. The tristimulus colorimeter is an instrument that measures a perceived color and the DRS instrument measures biological chromophores of the skin, including oxy- and deoxyhemoglobin, melanin and scattering. Data gathered from these too...

Using Raman Spectroscopy to Detect Malignant Changes in Tissues

Introduction Accurate, rapid and non-invasive detection and diagnosis of malignant disease in tissues is an important goal of biomedical research. Optical methods, such as diffuse reflectance, fluorescence spectroscopy, and Raman spectroscopy, have all been investigated as ways to attain this goal. Diffuse reflectance utilizes the absorption and scattering properties of tissues, particularly fr...

Detection and Characterization of Human Teeth Caries Using 2D Correlation Raman Spectroscopy

Background: Carious lesions are formed by a complex process of chemical interaction between dental enamel and its environment. They can cause cavities and pain, and are expensive to fix. It is hard to characterize in vivo as a result of environment factors and remineralization by ions in the oral cavity. Objectives: The development of a technique that gives early diagnosis which is non-invasi...

A Review of Surface-Enhanced Raman Spectroscopy on Potential Clinical Applications Towards Diagnosing Colorectal Cancer

Colorectal cancer (CRC) is one of the leading cancers in the world and early-screening is still the best method of cancer patient survival. However, colonoscopy as the current gold standard is not without flaws and an emerging technique called surface-enhanced Raman spectroscopy (SERS) coupled with machine learning is a possible candidate that could be applied in parallel with colonoscopy. This...
